In Vitro: Befovacimab binds human TFPI with high affinity (<10 pM) via the K1 ａnd K2 domains, inhibiting its interactions with both FXa ａnd FVIIa ａnd thus restoring their activity.

In Vivo: In monkey plasma, Befovacimab exhibits an IC₅₀ of 4.65 nM ａnd 6.19 nM for free TFPI ａnd diluted prothrombin time (dPT), respectively.
Befovacimab (5 ａnd 20 mg/kg; intravenous ａnd subcutaneous administration) dosedependent decreases in TFPI concentration ａnd clotting time in female cynomolgus monkeys in vivo.
