柱层析技术也称柱色谱技术。一根柱子里先填充不溶性基质形成固定相，将样品加到柱子上后用特别的溶剂洗脱，溶剂组成流动相。在样品从柱子上洗脱下来的过程中，根据混合物中各组分在固定向和流动相中的分配系数不同经过多次反复分配，将不同组分逐一分离。硅胶层析法的分离原理是根据物质在硅胶上的吸附力不同而得到分离，一般情况下极性较大的物质易被硅胶吸附，极性较弱的物质不易被硅胶吸附，整个层析过程即是吸附、解吸、再吸附、再解吸过程。根据填充基质和样品分配交换原理不同，离子交换层析，凝胶过滤层析和亲和层析是三种分离混合物的经典层析技术。
