（1）要注意避免出现严重溶血。血红蛋白中含有血红素基团，其有类似过氧化物的活性，因此，在以HRP为标记酶的elisa试剂盒测定中，如血清标本中血红蛋白浓度较高，则其就很容易在温育过程中吸附于固相，从而与后面加入的HRP底物反应显色。
（2）样本的采集及血清分离中要注意尽量避免细菌污染，一则细菌的生长，其所分泌的一些酶可能会对抗原抗体等蛋白产生分解作用；二则一些细菌的内源性酶如大肠杆菌的β-半乳糖苷酶本身会对用相应酶作标记的测定方法产生非特异性干扰。
（4）冰冻保存的血清标本须注意避免因停电等造成的反复冻融。ELISA试剂盒标本的反复冻融所产生的机械剪切力将对标本中的蛋白等分子产生破坏作用，从而引起假阴性结果。此外，冻融标本的混匀亦应注意，不要进行剧烈振荡，反复颠倒混匀即可。
（5）标本在保存中如出现细菌污染所致的混浊或絮状物时，应离心沉淀后取上清检测。Elisa试验是一种敏感性高，特异性强，重复性好的实验诊断方法。由于其试剂稳定、易保存，操作简便，结果判断较客观等因素，已广泛应用在免疫学检验的各领域中。ELISA试剂盒是用于体外定性检测人血清或血浆中的抗人类戊型肝炎（HEV）病毒 IgM 抗体ELISA检测。
