Target

• FunctionComponent of the Mediator complex, a coactivator involved in regulated gene transcription of nearly all RNA polymerase II-dependent genes. Mediator functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. Mediator is recruited to promoters by direct interactions with regulatory proteins ａnd serves as a scaffold for the assembly of a functional preinitiation complex with RNA polymerase II ａnd the general transcription factors. Phosphorylates the CTD （C-terminal domain） of the large subunit of RNA polymerase II （RNAp II）, which may inhibit the formation of a transcription initiation complex. Phosphorylates CCNH leading to down-regulation of the TFIIH complex ａnd transcriptional repression. Recruited through interaction with MAML1 to hyperphosphorylate the intracellular domain of NOTCH, leading to its degradation.

  Flow Cytometry - Anti-Cdk8 antibody （ab54561）
  Overlay histogram showing HeLa cells stained with ab54561 （red line）. The cells were fixed with 80% methanol （5 min） ａnd then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody （ab54561, 1μg/1x10⁶ cells） for 30 min at 22°C. The secondary antibody used was Alexa Fluor® 488 goat anti-mouse IgG （H&L） （ab150113） at 1/2000 dilution for 30 min at 22°C. Isotype control antibody （black line） was mouse IgG1 [ICIGG1] （ab91353, 1μg/1x10⁶ cells） used under the same conditions. Unlabelled sample （blue line） was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser （488nm） ａnd 525/30 bandpass filter.
