Target

• FunctionReceptor involved in the costimulatory signal essential for T-lymphocyte proliferation ａnd interleukin-2 production, by binding CD28 or CTLA-4. May play a critical role in the early events of T-cell activation ａnd costimulation of naive T-cells, such as deciding between immunity ａnd anergy that is made by T-cells within 24 hours after activation. Isoform 2 interferes with the formation of CD86 clusters, ａnd thus acts as a negative regulator of T-cell activation.

  ab53004, at a 1/100 dilution, staining Human CD86 in Tonsil, using Immunohistochemistry, Paraffin embedded tissue.

  Flow Cytometry - Anti-CD86 antibody [EP1158Y] （ab53004）
  Overlay histogram showing Raji cells stained with ab53004 （red line）. The cells were fixed with 4% paraformaldehyde （10 min） ａnd incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody （ab53004, 1/100 dilution） for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-rabbit IgG （H+L） （ab96899） at 1/500 dilution for 30 min at 22ºC. Isotype control antibody （black line） was rabbit IgG （monoclonal） （1µg/1x10⁶ cells） used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive result in 80% methanol （5 min） fixed Raji cells used under the same conditions.
