This protein amplifies neutrophil ａnd monocyte-mediated inflammatory responses triggered by bacterial ａnd fungal infections by stimulating release of pro-inflammatory chemokines ａnd cytokines, as well as increased surface expression of cell activation markers. Alternatively spliced transcript variants encoding different isoforms have been noted for this gene.[provided by RefSeq, Jun 2011].Function : Stimulates neutrophil ａnd monocyte-mediated inflammatory responses. Amplifier of inflammatory responses that are triggered by bacterial ａnd fungal infections ａnd is a crucial mediator of septic shock.Subunit : Interacts with TYROBP/DAP12.Subcellular Location : Isoform 1: Cell membrane; Single-pass type I membrane protein （Potential）. Isoform 2 was detected in the lung, liver ａnd mature monocytes.Similarity : Contains 1 Ig-like V-type （immunoglobulin-like） domain.TREM-1是近年来发现应用于诊断炎症性疾病的重要标记物,也是感染性疾病的预警指标,与细菌感染的严重程度相关。TREM-1出现早,半衰期短,在早期炎症诊断中很有意义。TREM 1属于免疫球蛋白超家族TREM,表达于白细胞的细胞膜表面受体,TREM-1选择性地表达于中性粒细胞和部分单核细胞, 它介导的信号传导通路在炎症的发生和级联放大中起重要作用。TREM-1激活后可上调TNF-α和IL-1等细胞因子的表达,并抑制抗炎症反应因子IL-10的表达,同时它激活的细胞因子又能上调TREM-1的表达。TREM-1与这些细胞因子之间形成的自分泌调节回路,促使炎症反应增强、放大。TREM-1能通过跨膜调节蛋白促发中性粒细胞和单核细胞的炎症反应,诱导炎症因子的分泌及钙离子总动员,使机体产生炎症并形成放大效应,使炎症反应弥散,甚至失去控制。
