Protein content ａnd activity of magnesium chelatase ａnd Rubisco:
Magnesium chelatase ａnd Rubisco were prepared by first freezing a 0.2-g leaf sample in liquid nitrogen to prevent proteolytic activity, followed by grinding with 2 mL of extraction buffer (100 mM phosphate buffer, pH 7.2, containing 0.1 mM EDTA, ａnd 2 mM ascorbic acid). The homogenate was centrifuged for 15 min at 10,000 rpm, ａnd the supernatant was used to measure the protein content (ng L–1), magnesium chelatase activity [pmol(Mg-Deutero) min–1 mg–1(protein)] ａnd Rubisco activity [μmol min–1 mg–1(protein)]. Magnesium chelatase ａnd Rubisco were measured using a magnesium chelatase content kit ａnd a Rubisco content kit (double antibody sandwich method, Huding Biological Technology Co., Shanghai, China) (3 replicates per treatment).
