人缺氧诱导因子-1α(HIF-1α)elisa试剂盒等ELISA试剂盒操作技术相关的注意事项:
⑴ 严格按照人缺氧诱导因子-1α(HIF-1α)elisa试剂盒说明书进行操作。操作前将试剂在室温下平衡30~60min。
⑵ 加样后及时放人孵箱。标本较多时,要分批操作。按说明步骤严格控制操作时间,防止孵育时间人为延长,导致非特异性结合紧附于反应孔周围,难以清洗彻底。
(3)封板温育时,各孔一定要封严,防止阳性标本的液体蒸发,产生周边现象从而导致“花板”的出现。
(4)用洗板机洗板时,保证洗液注满各孔,洗板针畅通,洗完板后最好在吸水纸(选择干净、无或少尘的吸水材料)上轻轻拍干:还要防止针口有纤维蛋白或异物,这些异物在洗板的过程中易产生拖带现象而导致“花板”的出现。
(5)合理安排检测量,以免反应板过多造成洗板等待时间长。
(6)加酶试剂后用吸水纸在酶标板表面轻拭吸干。
(7)显色剂尽量在临用前配制,坚持不用过期显色剂,肉眼可见浅蓝色的TMB显色剂不用。
(8)加样时保持显色剂不外流;A、B液应避免接触金属器械。加终止液时应避免产生气泡。
(9)应保证C清洁,整个操作过程中保证酶标板不接触次氯酸。以上的措施可以使“花板”降至最低限度。从而提高检测的特异性。并得到更准确、可靠的实验结果。

【应用原理】:
1.MASTER-10α是低浓度用产品 。
2.MASTER-10α产品带自动温度补偿功能及防水功能。
3.手持折射仪又叫手持折光仪,是能测定透明、半透明液体或固体的折射率nD和平均色散nF-nC的仪器(其中以测透明液体为主)。
4.折射率和平均色散是物质的重要光学常数,能借以了解物质的光学性能、纯度、浓度及色散大小等。仪器能测出蔗糖溶液内含糖量浓度百分数。
5.此种仪器使用范围甚广,是石油工业、油脂工业、制漆工业、食品工业、日用化学工业、制糖工业和地质勘察等有关工厂、学校及研究单位不可缺少的常用设备。
