Abstract:Dental materials are susceptible to dental plaque formation, which increases the risk of biofilm-associated oral diseases. Physical-chemical properties of dental material surfaces can affect salivary pellicle formation and bacteria attachment, but relationships between these properties have been understudied. We aimed to assess the effects of surface properties and adsorbed salivary pellicle on Streptococcus gordonii adhesion to traditional dental materials. Adsorption of salivary pellicle from one donor on gold, stainless steel, alumina and zirconia was monitored with a quartz crystal microbalance with dissipation monitoring (QCM-D). Surfaces were characterized by X-ray photoelectron spectroscopy, atomic force microscopy and water contact angles measurement before and after pellicle adsorption. Visualization and quantification of Live/Dead stained bacteria and scanning electron microscopy were used to study S. gordonii attachment to materials with and without pellicle. The work of adhesion between surfaces and bacteria was also determined. Adsorption kinetics and the final thickness of pellicle formed on the four materials were similar. Pellicle deposition on all materials increased surface hydrophilicity, surface energy and work of adhesion with bacteria. Surfaces with pellicle had significantly more attached bacteria than surfaces without pellicle, but the physical-chemical properties of the dental material did not significantly alter bacteria attachment. Our findings suggested that the critical factor increasing S. gordonii attachment was the salivary pellicle formed on dental materials. This is attributed to increased work of adhesion between bacteria and substrates with pellicle. New dental materials should be designed for controlling bacteria attachment by tuning thickness, composition and structure of the adsorbed salivary pellicle.

Journal:ACS Applied Materials & Interfaces

DOI: 10.1021/acsami.9b21405

Abstract:Total internal reflection microscopy (TIRM) is used to directly, sensitively, and simultaneously measure colloidal interactions, dynamics, and deposition for a broad range of polymer-surfactant compositions. A deposition state diagram containing comprehensive information about particle interactions, trajectories, and deposition behavior is obtained for polymer-surfactant compositions covering four decades in both polymer and surfactant concentrations. Bulk polymer-surfactant phase behavior and surface properties are characterized to provide additional information to interpret mechanisms. Materials investigated include cationic acrylamide-acrylamidopropyltrimonium (AAC) copolymer, sodium lauryl ether sulfate (SLES) surfactant, silica colloids, and glass microscope slides. Measured colloid-substrate interaction potentials and deposition behavior show non-monotonic trends vs. polymer-surfactant composition and appear to be synergistic in the sense that they are not easily explained as the superposition of single component mediated interactions. Broad findings show at some compositions polymer-surfactant complexes mediate bridging and depletion attraction that promote colloidal deposition, whereas, other compositions produce electrosteric repulsion that deters colloidal deposition. These findings illustrate mechanisms underlying colloid-surface interactions in polymer-surfactant mixtures, which is important to controlling selective colloidal deposition in multi-component formulation applications.

Journal:Chemical Engineering Science

DOI: 10.1016/j.ces.2020.115620

Abstract:Polymer inclusion membrane (PIM) has been progressively utilised in various chemical engineering applications. Understanding the interfacial interactions between PIM surface and surrounding solutions is significantly important for the industrial application of PIM systems. In this work, interfacial behavior of the PIM was studied during Zn(II) extraction process. The real-time change of the PIM under various environmental conditions was monitored by Quartz crystal microbalance with dissipation monitoring. Water penetration into the PIM layer was demonstrated when the PIM was in contact with Milli-Q water. Atomic force microscopy imaging confirmed that the swollen PIM had a looser network and rougher surface, as compared to its original state. When switching the surrounding environment to ZnSO4 solution, higher Zn(II) concentration could induce microstructure change and more significant membrane swelling in the PIM layer. Our results provide valuable implications for the design of effective PIM systems towards practical applications in wastewater treatment and hydrometallurgy.

Abstract:The hydraulic fracturing technique remains essential to unlock fossil fuel from shale oil reservoirs. However, water imbibed by shale during hydraulic fracturing triggers environmental and technical challenges due to the low flowback water recovery. While it appears that the imbibition of fracturing fluid is a complex function of physico-chemical processes in particular capillary force which is associated with wettability of oil-brine-shale, the controlling factor(s) to govern the wettability is incomplete and the literature data in this context is missing. We thus measured the adsorption/desorption of asphaltenes on silica surface in the presence of brines using quartz crystal microbalance with dissipation (QCM-D). We detected zeta potential of asphaltene-brine and brine-silica systems and calculated the disjoining pressures of the asphaltene-brine-silica system in the case of different salinity. Moreover, we performed a geochemical study to quantify the variation of surface chemical species at asphaltene and silica surfaces with different pH values and used the chemical force microscope (CFM) method to quantify the effect of pH on intermolecular forces. Our results show that lowering salinity or raising pH reduced the adhesion force between asphaltene and silica surface. For example, at a pH value of 6.5, when the concentration of injected water is reduced from 1000 mM to 100 mM and 10 mM, the adhesion force decreased by approximately 58% and 66%, respectively. In addition, for the 100 mM NaCl solution, when the pH value increased from 4.5 to 6.5 and 9, the adhesion force decreased by approximately 56% and 87%, respectively. Decreased adhesion forces between asphaltene and the silica surface could promote the desorption of asphaltene from the silica surface, resulting in a negative zeta potential for both asphaltene-silica and brine-silica interfaces and a shift of wettability towards water-wet characteristic. During such a process, -NH+ number at asphaltene surfaces decreases and the bonds between -NH+ and >SiO− break down, to further interpret the formation of a thinner asphaltene adlayer on the rock surface. This study proposes a reliable theoretical basis for the application of hydraulic fracturing technology, and a facile and possible manipulation strategy to increase flowback water from unconventional reservoirs.

Journal:Journal of Membrane Science

DOI: 10.1016/j.memsci.2020.118046

Abstract:While polyvinylidene fluoride (PVDF) is widely used as a commercial UF membrane material, PVDF membrane often encounters fouling problems which significantly reduce the separation performance and service life of membrane. In this work, a special fluoro-contained polymer, sulfonated poly(α,β,β-trifluorostyrene) (SPTFS), is used to improve the hydrophilicity of PVDF membrane, thus to enhance the water flux and antifouling property of the membrane. PVDF membranes with different doping amount of SPTFS are prepared through conventional non-solvent induced phase separation method. Effects of doping SPTFS on the surface morphology, porosity, water contact angle and zeta potential of the prepared membranes are investigated. Compared with the pristine PVDF membrane, the SPTFS-doped membranes show improved hydrophilic property with enlarged finger-like pore structure, decreased water contact angle and higher negative surface charge density, as well as the increased filtration water flux. The enhanced anti-fouling property of the membrane is demonstrated by the cyclic filtration test and the in-situ monitoring of the adsorption and desorption of pollutants using a quartz crystal microbalance.

Journal:European Journal of Inorganic Chemistry

DOI: 10.1002/ejic.202000096

Abstract:A high demand for materials with defined geometry and size is required in a wide range of fields. Inorganic compounds, especially silica‐based, arise as a cheap source and chemically flexible for the purpose. Silica display unique properties, like easy functionalization and good optical features making an interesting material to manipulate. In this work, we developed a method to create thin silica microsheets with defined size and high‐fidelity shape using superhydrophobic‐hydrophilic microarrays. These microstructures were produced through sol‐gel process using biomimetic superhydrophobic surfaces decorated with wettable spots. The results confirm the manufacture of porous silica microstructures with defined design (squares and circles) and thickness around 7 µm. The methodology applied in this work enables the high throughput fabrication of shaped silica materials in a single step, unlocking an extensive number of applications in areas that require miniaturization, like microelectronics or in fields like sensing and biomedicine.

7. Name:Chitosan/Alginate Dialdehyde Multilayer Films with Modulated pH‐Responsiveness and Swelling

Authors:Junhao Huang, Saeed Zajforoushan Moghaddam and Esben Thormann

Journal:Macromolecular Chemistry and Physics

DOI: 10.1002/macp.201900499

Abstract:Oxidation of alginate is a promising method that creates aldehyde groups, which can form stable cross‐links with amine groups on chitosan. In this work, it is shown that a systematic variation in the oxidation degree of alginate dialdehyde allows for effective tuning of the swelling capacity of chitosan/alginate dialdehyde multilayer films. Furthermore, it is demonstrated that the pH‐responsiveness of the multilayer films can be regulated by the choice of pH at which the layer‐by‐layer assembly is conducted. The combination of these two parameters allows for the designing polysaccharide multilayer films with a tunable degree of swelling and stimuli‐responsiveness.

Journal:International Journal of Molecular Sciences

DOI: 10.3390/ijms21062211

Abstract:The compound 1,1,1-trichloro-2-(p-chlorophenyl)-2-(o-chlorophenyl) ethane (o,p’-DDT) has been identified as one of the endocrine-disrupting chemicals causing adverse effects on wildlife and even humans through bioaccumulation. Its detection has become increasingly important. We have obtained candidate aptamers binding to o,p’-DDT by a systematic evolution of ligands by exponential enrichment (SELEX) protocol. Five out of seventeen candidate sequences were selected for preliminary characterization by SYBR Green I assay. One sequence with highest fluorescence response with o,p’-DDT, designated DDT_13, was chosen for further characterization. Its dissociation constant (Kd) was determined to be 412.3 ± 124.6 nM. DDT_13 exhibited low cross-binding activities on other tested small molecules. The good bioactivities of DDT_13 were demonstrated for the analysis of spiked lake water and tap water samples. This study provides a novel o,p’-DDT-specific probe for its future applications.

Journal:Chalmers University of Technology, Chemistry and Chemical Engineering

DOI:

Abstract:Capture and release of proteins by an electrical signal is of general interest but methods have proven difficult to develop. We present a polyelectrolyte brush electrode with high-capacity (several μg/cm2) for protein immobilization that captures and releases proteins by an electrochemical potential. The electrochemical potential produces an interfacial pH gradient that enables rapid and tunable switch of the interaction between the polyelectrolyte brush and proteins on-demand. Key to our solution is an electrochemically inert aryl anchor between the polyelectrolytes and the electrode, which enables multiple capture-release cycles (without any regeneration step) and minimal degradation of the electrode. For all proteins tested we observed complete release by electrochemistry. In addition to proteins, the electrode could successfully immobilize and release liposomes suggesting additional applicability within the field of biomedicine and drug delivery. Importantly the electrode retained its functions when exposed to serum suggesting it can be used in a biological environment. We predict that this technology will be highly useful for protein purification and in biomedical devices.

10. Name:Synergy between composite nanoparticles and saponin beta-escin to produce long-lasting foams

Authors:Kai Yu, Bin Li, Zhentao Wang, Wei Zhang, Dongbao Wang, Haojie Xu, Junfeng Wang and David Harbottle

Journal:Industrial & Engineering Chemistry Research

DOI: 10.1021/acs.iecr.9b06355

Abstract:A synergy between poly(vinylpyrrolidone) (PVP)-coated silica nanoparticles (CPs) and saponin beta-escin was exploited to develop long-lasting foams using low component concentrations. Individually, foams stabilized by either CPs or saponin beta-escin suffered from bubble coarsening and bubble coalescence, respectively. In contrast, the successful synergy of CPs and saponin beta-escin was shown to provide excellent foamability and foam stability, with no direct interaction between the two components. The CPs aggregated strongly in 0.55 M Na2SO4 and formed a porous stabilizing network, hindering bubble coalescence but not bubble coarsening. However, foams stabilized by saponin beta-escin and CPs, the bubble interface was saturated by the two components, with CPs forming thick liquid lamellae between bubbles, hindering liquid drainage and bubble coalescence, and saponin beta-escin (added at 0.01 wt%) saturating the pore-space in the CP interfacial network to retard bubble coarsening. Adding saponin beta-escin (added at 0.01 wt%) as a co-foaming agent, the foamability and foam lifetime were increased by ~2.5 and ~4 times, respectively, compared to CP-stabilized foams.

11. Name:Impact of antigen density on recognition by monoclonal antibodies

Authors:Laure Bar, Jérôme Dejeu, Rémy Lartia, Fouzia Bano, Ralf P. Richter, Liliane Coche-Guérente and Didier Boturyn

Journal:Analytical Chemistry

DOI: 10.1021/acs.analchem.0c00092

Abstract:Understanding antigen–antibody interactions is important to many emerging medical and bioanalytical applications. In particular, the levels of antigen expression at the cell surface may determine antibody-mediated cell death. This parameter has a clear effect on outcome in patients undergoing immunotherapy. In this context, CD20 which is expressed in the membrane of B cells has received significant attention as target for immunotherapy of leukemia and lymphoma using the monoclonal antibody rituximab. To systematically study the impact of CD20 density on antibody recognition, we designed self-assembled monolayers that display tunable CD20 epitope densities. For this purpose, we developed in situ click chemistry to functionalize SPR sensor chips. We find that the rituximab binding affinity depends sensitively and nonmonotonously on CD20 surface density. Strongest binding, with an equilibrium dissociation constant (KD = 32 nM) close to values previously reported from in vitro analysis with B cells (apparent KD between 5 and 19 nM), was obtained for an average inter-antigen spacing of 2 nm. This distance is required for improving rituximab recognition, and in agreement with the known requirement of CD20 to form clusters to elicit a biological response. More generally, this study offers an interesting outlook in the understanding of the necessity of epitope clusters for effective mAb recognition.

12. Name:Swelling behavior, interaction, and electrostatic properties of chitosan/alginate dialdehyde multilayer films with different outermost layer

Authors:Junhao Huang, Saeed Zajforoushan Moghaddam, Plinio Maroni and Esben Thormann

Journal:Langmuir

DOI: 10.1021/acs.langmuir.0c00330

Abstract:In this study, self-cross-linked chitosan/alginate dialdehyde multilayer films, capped with either alginate dialdehyde (6 layers) or chitosan (7 layers), are fabricated using the layer-by-layer method. The disruption of the electrostatic equilibrium when exposing the fabricated layers to acidic and alkaline conditions causes swelling within the film and independently in the outermost layer, showing dependence on the ionic strength. Spectroscopic ellipsometry and quartz crystal microbalance with dissipation monitoring are employed to examine the swelling behavior. Atomic force microscopy colloidal probe measurements are conducted to assess the surface forces between the multilayer films at different pH and ionic strengths. Finally, the electrostatic properties of the multilayer films are examined at different pH and ionic strengths using zeta potential measurements. The results suggest that stimuli-responsiveness and overall swelling behavior of the polysaccharide multilayer films significantly depend on the outermost layer, an effect that should expectedly become more pronounced the thinner the film becomes.

13. Name:Spontaneous formation of cushioned model membranes promoted by an intrinsically disordered protein

Authors:Yuri Gerelli, Amanda Eriksson Skog, Stephanie Jephthah, Rebecca J. L. Welbourn, Alexey Klechikov and Marie Skepö

Journal:Langmuir

DOI: 10.1021/acs.langmuir.0c00120

Abstract:In this manuscript it is shown that by exposing commonly used lipids for bio- membrane mimicking studies to a solution containing the histidine rich intrinsically disordered protein Histatin 5, a protein cushion spontaneously forms underneath the bilayer. The underlying mechanism is attributed to have an electrostatic origin, and it is hypothesized that the observed behaviour is due to proton charge fluctuations promoting attractive electrostatic interactions between the positively charged proteins and the anionic surfaces, with concomitant counterion release. Hence, we anticipate that this novel "green" approach of forming cushioned bilayers can be an important tool to mimic the cell membrane without the disturbance of the solid substrate, and thereby achieving a further understanding of protein-cell interactions.

14. Name:New unsymmetrical bisacridine derivatives non-covalently attached to quaternary quantum dots improve cancer therapy via enhancing cytotoxicity towards cancer cells and protecting normal cells

Journal:ACS Applied Materials & Interfaces

DOI: 10.1021/acsami.0c02621

Abstract:The use of nanoparticles for the controlled drug delivery to cells has emerged as a good alternative to traditional systemic delivery. Quantum dots (QDs) offer potentially invaluable societal benefits such as drug targeting and in vivo biomedical imaging. In contrast, QDs may also pose risks to human health and the environment under certain conditions. Here, we demonstrated that unique combination of nanocrystals core components (Ag-In-Zn-S) would eliminate the toxicity problem and increases their biomedical applications. The alloyed quaternary nanocrystals Ag-In-Zn-S (QDgreen: Ag1.0In1.2Zn5.6S9.4 and QDred: Ag1.0In1.0Zn1.0S3.5) were used to transport new unsymmetrical bisacridine derivatives (UAs: C-2028 and C-2045) into lung H460 and colon HCT116 cancer cells for improving the cytotoxic and antitumor action of these compounds. UAs were coupled with QD through physical adsorption. The obtained results clearly indicate that the synthesized nanoconjugates exhibited higher cytotoxic activity than unbound compounds, especially towards lung H460 cancer cells. Importantly, unsymmetrical bisacridines non-covalently attached to QD strongly protect normal cells from the drug action. It is worth to point out that QDgreen or QDred without UAs did not influence the growth of cancer as well as normal cells what is consistent with in vivo results. In non-cellular systems, at pH 5.5 and 4.0, which relates to the conditions of endosomes and lysosomes, the UAs released from QD-UAs nanoconjugates. An increase of total lysosomes content was observed in H460 cells treated with QDs-UAs what can affect the release of the UAs from the conjugates. Moreover, confocal laser scanning microscopy analyses revealed that QD-UAs nanoconjugates enter to H460 cells more efficiently than to HCT116 and normal cells, which may be the reason of their higher cytotoxicity against lung cancer. Summarizing, the non-covalent attachment of UAs to QDs increases the therapeutic efficiency of UAs, by improving cytotoxicity toward lung H460 cancer cells and having protecting effects on normal cells.
