一、Deｓcription：

The Origami B host strain carries the same mutations as the original Origami strain in trxB and gor, but they originate from the BL21 lacZY mutant.

The expression levels can be precisely controlled by adjusting IPTG concentration.

Thus, Origami B integrates the ideal characteristics of BL21, Tuner, and Origami strains within a single bacterial background.

The mutations in trxB and gor are selectable under kanamycin and tetracycline respectively, meaning these strains cannot be used with plasmids that require either kanamycin or tetracycline for selection.

Additionally, these strains contain BL21's lon and ompt deficiencies, which enhance protein stability.

DE3 indicates that the host is a λDE3-competent lysogen, thus carrying chromosomal copies of the T7 RNA polymerase gene under the control of the lacUV5 promoter.

This strain is suitable for inducing protein production by expressing the target gene cloned in the pET vector using IPTG.

Origami B宿主菌株在trxB和gor中携带与原始Origami菌株相同的突变，只是它们来自BL21的lacZY突变体，可以通过调节IPTG的浓度来控制表达水平。

因此Origami B菌株在一个菌株背景中结合了BL21、Tuner和Origami菌株的理想特征。

trxB和gor的突变分别在卡那霉素和四环素上是可选择的；因此，这些菌株不能与只能用卡那霉素或四环素选择的质粒一起使用。

这些菌株还包括BL21的lon和ompT缺陷，可增加蛋白质的稳定性。

二、使用场景：

DNA化学转化：用于外源质粒DNA（如pET、pGEX、pMAL系列载体）的热击转化实验，转化效率可达10⁷–10⁸ cfu/μg DNA（以pUC19为对照质粒）。

蛋白质表达：

特别适用于表达含二硫键的难溶性蛋白，因其基因型包含突变的硫氧还蛋白还原酶（trxB）和谷胱甘肽还原酶（gor），可促进二硫键正确折叠，提高蛋白可溶性。

整合λ噬菌体DE3溶原体，表达T7 RNA聚合酶，需配合IPTG诱导（适用于T7启动子载体，如pET系列）。

抗性限制：

菌株天然具有卡那霉素（KanR）和四环素（TetR）抗性，因此不能用于携带这两种抗性的质粒表达（如pUC19对照实验需避开KanR/TetR抗性质粒）。

适用于氨苄青霉素（AmpR）等非Kan/Tet抗性的质粒。
