一、基本描述：

CopyCutter Induction Solution是一种用于动态调控大肠杆菌质粒拷贝数的分子生物学试剂。

其核心设计目的是解决不稳定DNA序列或毒性基因的克隆难题，同时兼顾质粒产量的提升。

作用对象：

EPI400菌株：通过改造宿主菌的基因实现调控。

该菌株删除了组成型表达的pcnB基因，替换为诱导型启动子控制的改良pcnB基因。

未诱导时质粒维持低拷贝，加入诱导液后激活表达，显着提高质粒产量。

EPI300菌株：诱导液激活突变的trfA基因，促进含oriV复制子的质粒高拷贝扩增。

物理特性：

以无菌过滤的浓缩液形式提供（常见200X或1000X浓度），可直接加入培养基使用。

保存条件：-20℃长期稳定（≥2年），4℃保存限6个月；若浑浊需用0.2-0.45μm滤膜过滤。

CopyCutter Induction Solution 使用优点：

（1）CopyControl pCC1FOS和pCC2FOS裁体：线性化,去磷酸化,纯化,已经为连接做好准备。

（2）不需要限制性内切酶消化或脉冲凝胶电泳来准备用于克隆的基因组DNA。

（3）使用pCC2FOS裁体可以最大化高通量末端测序结果。

（4）可以诱导单拷贝到每个细胞50拷贝。安全地获得较高的DNA产量,同时保持了稳定克隆单拷贝数。

（5）高效率的lambda包装蛋白消除了背景值假阳性。

（6）比BAC克隆更快更容易。

二、注意事项Notes:

（1）Competent cells must be transported on dry ice. Competent cells should be stored at -80C, and should not be repeatedly freeze-thawed and left for too long, otherwise the transｆormation efficiency of competent cells will be reduced.

（2）It is best to melt competent cells on ice.

（3）When perｆorming transｆormation operations, it should be carried out at the corresponding temperature and sterile conditions.

（4）Transｆorming high concentrations of plasmids correspondingly reduces the amount of bacteria that end up being used for coating plates.

（5）To prevent unsuccessful conversion experiments, part of the junction reaction solution can be retained for reconversion to minimize losses.

（6）The product package contains the vector pUC19 DNA (10pg/ul) for control experiments.

（7）For your safety and health, please wear lab coats and disposable gloves for operation.
