Description
The detection of proteins in immunoblotting applications is typically achieved by using an antibody that is conjugated to an enzyme, such as horseradish peroxidase (HRP) or alkaline phosphatase (AP). The primary antibody which directly binds to the protein target may be labeled with the enzyme, or a labeled secondary antibody may be used to bind to an unlabeled primary antibody. The covalently attached enzyme is capable of degrading a substrate, which may be chromogenic or chemiluminescent. Chromogenic substrates are degraded into colored precipitates ａnd are easy to use. No special equipment is required for detection. Chemiluminescent reactions produce light ａnd are more sensitive than chromogenic reactions, but the image must be recorded on x-ray film or with a gel documentation system. For both types of substrates, the signal generated by the enzyme catalyzed reaction is detected on the membrane only at the site of the target protein, if the immunoblotting conditions are optimized.
AMRESCO＇s Western Max™ Chemiluminescent Kits contain all the necessary components to obtain sensitive detection of proteins, except for the primary antibody. The kits are compatible with both PVDF ａnd nitrocellulose. Kit includes IgG Positive Control, Dilution Buffer Powder, Tween® 20, BSA, Fraction V, Western Max™ Guide with Protocol. Each kit has enough reagents for forty 10 cm×10 cm immunoblots.
