用特异性抗体包被于固相载体，经洗涤后加入含有抗原之待测样品，如待检样品中有相应抗原存在，即可与包被于固相载体上的特异性抗体结合，经保温孵育洗涤后，即可加入酶标记特异性抗体，再经孵育洗涤后，加底物显色进行测定，底物降解的量即为欲测抗原的量。这种方法欲测的抗原必须有两个可以与抗体结合的部位，因为其一端要包被于固相载体上的抗体作用，而另一端则要与酶标记特异性抗体作用。因此，不能用于分子量小于5000的半抗原之类的抗原测定。

可以：（1）免疫酶染色各种细胞内成份的定位。

（2）研究抗酶抗体的合成。

（3）显现微量的免疫沉淀反应。

（4）定量检测体液中抗原或抗体成份。

血浆样本直接测定前处理方法：

（1）加酶抑制剂：准确采全血若干毫升，注入预冷的加有抗凝剂的塑料指形管中，迅速低温离心，取血浆低温保存待测。抑肽酶有液体的与固体的。固体的抑肽酶可用生理盐水溶解。

（2）酸化处理：每毫升血浆加1mol/l HCl 0.2ml，低温保存待测。以上两种方法，任选一种即可。

血浆样本的间接测定 血标本经提取后，可去除蛋白质等干扰因素，并使微量的生物活性肽浓缩，但较费时，成本也高。常用的提取方法有：

（1）柱层析提取法：有多种层析柱可供提取不同的神经肽，其中较为方便的是用Sep-pak C18层析柱提取。主要步骤：

①柱的预处理：该柱有两头，长头连接注射器，可注入溶液与样品，短头为排出口。预处理时注入乙腈10ml。蒸馏水20ml，使柱中的生物胶活化。

②注入样品（如血浆、脑脊液、腹水、尿等）：样品中的水份流出，生物活性肽、蛋白及杂质等吸附在生物胶上。血浆上柱前，如先去除蛋白，可增加柱子的使用时间。

③淋洗：用4%HAC溶液100ml注入柱中，以洗去一些杂质。

④洗脱：用7ml酸性乙腈作为洗脱剂，注入柱中，把生物活性肽洗脱下来，收集在塑料指形管中。

⑤清洗：用乙腈、蒸馏水清洗柱子，以备后用。

⑥将洗脱液吹干，低温保存待测。测定时可加入一定量的缓冲液溶解。
