测定OD值;10、阳性OD值比对照值大3倍,为判断效价稀释度。多肽的溶解与保存:多肽有着很多特性,关于一些特殊肽您需要的有关信息可与博奥森公司的技术人员联系。当然首先建议您用少量肽来试验适合的溶解方法,当多肽完全溶解后,才能加缓冲液(注意:应将多肽加入到适当溶剂中搅拌)。一、冻干多肽的溶解1、多肽溶解的主要问题是二级结构的形成。虽然疏水肽链二级结构的形成更明显,但除短的肽链外,此现象产生在几乎所有肽链,与极性无关。所以,溶解多肽的个原则是使用无菌蒸馏水或去离子水,当然无氧水。多肽溶液可能遇到细菌降解,为防止此情况的发生,应溶解在无菌的蒸馏水中或用0.45或0.2孔径的滤膜过滤除菌。含有Cys、Met、Trp的多肽很容易氧化,应溶于无氧的水中,无氧水可通过注入惰性气体(氮、氦、氩)减压除气得到;2、若多肽不溶于纯水,超声处理有助于打碎颗粒并增加溶解度。注意:超声处理会引起溶液发热或多肽降解;3、若多肽含有多个碱性氨基酸,使用(1-10%)的乙酸水溶液:对于疏水性强的多肽,应使用50%的乙酸;4、若多肽中含有大量酸性氨基酸,可用氨溶液(1-10%)或乙酸乙基吗啡或重碳酸盐等挥发性的碱性缓冲液溶解。pH值在层析前必须调整;5、异丙醇和乙腈能溶解中等大小的多肽。若肽要上柱,有机溶剂的量必须很少否则将严重影响停留时间;6、若多肽因含有Val、Leu、Met、Phe、Tyr、Ala等芳香链而高度疏水、或为中性肽时,DMF或DMSO等膜变性剂的使用,有利于多肽的溶解:a.高浓度模变性剂通过破坏多肽的二级结构而助溶;b.膜变性剂适于多肽分析液的制备,但可能会对其生物活性的研究工作造成干扰;c.DMF是变性剂(浓度可达30%),滴加至多肽溶解;d.反相层析法时,DMF将与洗脱液前锋一起流出,根据入量的多少,峰值可能很高。大多数肽能在大量DMF流出后几分钟内流出,若肽链很小,洗脱太早,多肽的量将很低。二、冻干多肽的保存标有“冻干保存”说明的多肽应该保存在冰冻条件下,以-20℃以下,其活性可保持多年。当使用干冻产品时,开盖前其容器应在装有新鲜干燥箱内升至室温。-20℃的产品,此过程需要一小时或更长,随保装而定。否则,当容器打开,水气进入导致凝缩而降低稳定性。一旦打开,应迅速称量完毕,立即封闭以免潮解,亲水肽更应注意。三、溶解多肽的保存多肽溶液比干粉的稳定性差很多,为了获得较好的效果,遵循以下原则:1、分成小包装避免反复冻融。使用多少解冻多少;2、溶于pH5-7无菌的缓冲液中,-20℃储存;3、由于细菌能降解多肽,储存前应过滤细菌;4、包含Cys、Met、Try、Glu、Asp的多肽易于氧化,因此应保存在无氧化剂的环境中。我公司免费提供抗血清亲合层析纯化服务,得到目的蛋白的抗体IgG。我公司还免费提供真空冷冻抽干服务,得到冻干粉状的抗体IgG产品,以便产品的使用及长期保存。制备要求:客户需提供制备抗体的相关蛋白的英文名称的全称,也可同时提供该蛋白的全序列或氨基酸(多肽片断)序列,供参考(不要求)
